Comparison between Germinated Seed and Isolated Microspore EMS Mutagenesis in Chinese Cabbage (Brassica rapa L. ssp. pekinensis)

Mutagenesis is an important tool for breeding and genomic research. In this study, the germinated seeds isolated microspores of a double haploid line ‘FT’ were treated with EMS, respectively, aim comparing effects two approaches on generating mutants in Chinese cabbage. For microspore EMS mutagenesis, 0.12% 20 min, total 1268 plantlets obtained, 15 M1 screened mutation frequency 1.2%. seed 7800 0.8% 12 h, 701 M2 screened, 18.78%. total, 716 heritable morphological variation including leaf color, shape, leafy head, bolting, fertility, obtained from mutagenesis experiments. Homozygous mutant plants could be lines by mutagenesis. The was higher than Based these results, we propose that more suitable to generate large-scale library, conducive rapidly obtaining homozygous mutants.
